First, Great that you are back and are Working on the Map again. Now to your latest map and the 2 Heroes(Vampire and Life Manipulator).
Vampire
Q-Ability: in the early stages of the Game, the 50% hp will 99,99% kill you, but you will 100% kill everything in you Range. In the Lategame(30+), where you have lots of Str thx to WeaponCraft and Items, the 50% is way too much of a loss for a pretty weak spell itself. If i hadn't a Life Manipulator on my Side, i would have never used the Spell lategame. You should consider doing something with it.
W-Ability: Can stay exactly the way it is, it just suits him and works fine.
E-Passiv: The Idea behind it is good and early it does its job, but Lategame where you have 100-200k hp, you dont feel anything of it anymore.
Ultimate: The Current Ultimate is on this Style useless. you reduce 50% dmg taken, doesnt really matter because most of the dmg you get, you deal yourself, and the heal from enemy attacks is too low to give it a Function as an Ultimate. An Idea would be, to remove the 50% reduce thing and make it an Active, that last 5-6 seconds and instead of getting dmg from enemy hits, you heal yourself, with 90-120sec cd.
Omega: He just needs it and fits perfectly

Class Item: 250allstats...pretty low, then the effect that u drain 5% of target mana and do that amount in dmg...most enemies dont have mana, and if they have mana, its a low amount of mana that you dont feel the effect at all. Consider his Unique Mechanic a bit more, how about reducing the dmg your Q does to yourself to 25%-30%, that would be usefull as classitem.
Life Manipulator
Where to start...he brakes the game...i played with 1 friend, he had invoker, at round 10 we had 500 allstats, round 20 we had 2-3k allstats and that just snowballed further out of control after he received the eye, at dragon round we had over 22k all stats.
His Ultimate is a good idea and works fine, however plz remove buildings out of his targets. he can reduce the hp from buildings(you cant kill them, but shouldnt happen)
Class Item is way too strong, he can spam his Healcircle, 2sec cd and 12 sec duration(or something like that) is a joke, especially that it now heals buildings too. And then there is the point, that with his class item he has "4" necklaces(1. Powercraft x7, 2. Powercraft x7#2, 3. Spellcraft x7, 4. Grand necklace).
And in the Boss round Absolute Zero, the Legendary Item "Absolute Zero" spawns random around the Castle, why does a op legendary item spawn?
