The classes are...
Wizzard
Barbarian
Witch Doctor
Monk
Demon Hunter
More classes will be added for the expansions. There will probably be 2 expansions and we can expect 2 - 3 classes added to each.
Classes are a lot more customizable than in D2, with a large number of viable moves.
As for release date... A beta could be up as early as after blizzcon this year.
Actual release will probably be sometime nextyear. Depends on delays and problems they encounter.
