When i create a defeat condition on an existing keep. Will the trigger still work when the keep is upgraded to a castle?

First impression: yes, the building just got transformed.

Secondly: this is very easy to test, isn't it?
No offense btw, i just think you can make a blank map and try very quickly.

My thougths exactly. OP, it's absolutely not a problem that you've asked, but I'd strongly encourage you to experiment with things like these - you'll learn more that way

Anyway, the answer is yes, it will work.
