Well the full trigger is:

  • Untitled Trigger 001
    • Events
      • Unit - A unit Starts the effect of an ability
    • Conditions
      • (Ability being cast) Equal to Run
    • Actions
      • Trigger - Turn on Untitled Trigger 002 <gen>
  • Untitled Trigger 002
    • Events
      • Time - Every 1.00 seconds of game time
    • Conditions
    • Actions
      • Unit - Set mana of (Casting unit) to 0.00
What i want is so that the unit that casted run would run faster and for that his mana will be drained.
But i cant put a negative value for mana.

No. You don't need negative. There is the ability called Mindrot (ANmr). Neutral Hostrile --> Units.
You can cast it with dummy on hero when he uses a speed up ability.
And movement speed can be changed with triggers.

Superclaude, that behaviour is to be predicted.

Why are you trying to set your units mana to -1? Especially since it is impossiable for units to have negative mana, it is only logical that it should not let you.

I do believe you are after setting the unit's mana to (unit's current mana -1), afterall that sort of sounds like what you are after.

However, if you still want the unit's mana to be -1, use JASS as there are no number restrictions there. However, I would still recommend using 0 over -1 as they will both have the same effect.

Seems to me like he is just trying to use the mana pool as say, a stamina pool. IE Diablo style.

In that case, just do what someone else said set up a trigger and use a dummy unit to cast Mind Rot on your hero when you use the "Run" ability.
